It is readily absorbed from the GI tract, 99% bound to plasma protenins,
distributed into synovial fluid, the central nervous sytem, placenta and
breast milk. It is is metabolised in the liver to its glucuronide
conjugates, excretion of metabolites is predominantly in the urine with
some amount appearing in the faeces.
Oral
RA, OSTEOARTHRITIS, ANKYLOSING SPONDYLITIS, ACUTE GOUT ACUTE MUSCULO-SKELETAL
DISORDERS: 25mg 2 or 3 times daily. Oral Sust. Rel caps (75mg) in cap
o.d.
Action
Anti-inflammatory, analgesic, antipyretic and antigout actions.
Potent inhibitor of cyclo-oxyenase, reducing prostaglandin synthesis.
Relieves pain and reduces temperature in febrile patients. Reduces pain
and joint swelling in rheumatoid arthritis (RA) but does not modify
progress. Also used topically in the eye to reduce local inflammation.
Interactions
Antigoagulants: May prolong prothrombin time. Instruct patients to
watch for signs & symptoms of bleeding. ACE inhibitors:
Antihypertensive effects of captopril may be blunted or completely
abolished by indomethacin.
Beta-Blockers: Antihypertensive effect may be blunted.
Cyclosporine: Nephrotoxicity of both agents may be increased.
Digoxin: Serum digoxin levels may be increased by ibuprofen.
Adverse
Effect & Precautions
Nausea, vomting, anorexia, gastric bleeding, diarrhoea, dizziness,
frontal headache, confusion, depression, psyache, confusion,
depression, psychosis, hallucination and leukopenia, epigastric
distress. Rarely aplastic anaemia. Most adverse effects are
dose related.
Precaution: Impairment of hepatic function
Pregnancy: Contraindicated.
Breast Feeding: Contraindicated.
Man: Use with caution.
