Pharmacokinetics

      

Readily absorbed from GI
tract. Food slows the rate of absorption but not the total bio-availabilty.
Extensively bound to plasma proteins and substance concentrations are
found in synovial fluid. Metabolised mainly conjugation with glucuronic
acid and excreted mainly in the urine.

Indication
& Dosage


 



Oral

RHEUMATOID AND OSTEOARTHRITIS, ACUTE GOUT, CAPSULITIS OF THE
SHOULDER, PAINFUL MUSCULO-SKELETAL CONDITIONS:
50mg 2 or 3 times daily.
May be increased upto 300mg/day in severe cases.

MILD TO MODERATE PAIN AND DYSMENORRHOEA: 25-50mg 6-8 hrly.

Children: Not recommended.

Action

 


Ketoprofen is a propionic acid derivative with anti-inflammatory,
analgesic and antipyretic actions. Inhibits synthesis of prostaglandins
and leukotrienes. It also stabilises lysosomal membrances.

 

Interaction

 

Acetylsalicylic acid /  Plasma ketoprofen levels due to  plasma protein binding

Hydrochlorothiazide /  Chloride and potassium excretion


Methotrexate / Concomitant use  toxic plasma levels of methotrexate


Probenecid /  Plasma clearance of ketoprofen and  plasma protein binding


Warfarin / Additive effect to cause bleeding

Adverse Effect
& Precautions


 



Nausea, vomting, epigastric
discomfort, dizziness, headache, skin rash, thrombocytepenia.



Precaution: Bleeding disorders, aspirin or
anti-inflammatory drug induced allergy, asthma, hypertension, hepatic or
renal or cardiac dysfunction.

Preganany: Contraindicated

Breast Feeding: Contraindicated

Man: May be used in reduced dose
