Well absorbed by oral or
intramuscular administration and is rapidly converted to bilogically
active folated distribution occurs to all body tissues and is
concentrated in the CSF. It is excreted in the urine.
Indication
& Dosage
IM/IV
ANTIDOTE FOR METHOTREXATE TOCITIY: Amount equal to methotrexate dosage
administered as early as possible.
ADJUNCT THERAPY WIT 5 FU IN COLORECTAL CANCER: 200mg/mg by slow IV
injection over 3 mins followed by 5 FU 370mg/m2 IV. Daily for 5 days.
Enter body folate pool as 5-methyltetrahydro folate. Cna neutralise the effects of folic acid antagonists such as methotrexate but increases those of 5-FU.
Interactions
Methotrexate:
Methotrexate toxicity reduced.
Fluorouracil: Cytotoxic effects of fluorouracil enhanced
Folic acid antagonist: Should not be coadministered with a folic
acid antagonist.
Adverse
Effect & Precautions
Rarely
pyrexia.
Precaution: Should not be administered with folic acid antagonists
Pregnancy: Safety not established.
Breast Feeding: Safety not established.
