Lincomycin is given orally and intramuscularly. The drug is well distributed throughout the body and is mainly concentrated in the liver, spleen, kidney and lungs.Plasma half life is 5 hrs.
IM/IV
INFECTIONS WIHT SUSCEPTIBLE ORGANISMS WHICH ARE RESISTANT TO OHTER
ANTIBIOTICS. IN PATIENTS WHO ARE ALLERGIC TO PENICILLINS. TONSILLITIS,
PHARYNGITIS, OTITIS MEDIA, SINUSITIS, ACUTE AND CHRONIC BRONCHITIS,
PNEUMONIA, SKIN AND SOFT TISSUE AND BONE & JOINT INFECTIONS,
OSTEOMYELITIS, SEPTIC ARTHRITIS, SEPTICAEMIA, ENDOCARDITIS: 600mg IM once
or twice daily or IV infusion 8-12 hrly of 600mg-1g. Higher doses upto 8g
have been given in life-threatening infections. For IV infusion lincomycin
1g is diltuted in 100ml of a parenteral solution and given at the rate of
100ml/hour Chidlren: 10mg/kg IM once or twice daily or 10-20mg/kg/day as
IV infusion in divided doses every 8 or 12 hours.
Oral
INFECTION WITH SUSCEPTIBLE ORAGANISMS WHICH ARE RESISTANT TO OHTER
ANTIBIOTICS. IN PATIENTS WHO ARE ALLERGIC TO PENICILLINS. TONSILLITIS,
PHARYNGITIS, OTITIS MEDIA, SINUSTIS, ACUTE AND CHRONIC BRONCHITIS,
PNEUMONIA, SKIN AND SOFT TISSUE AND BONE & JOINT INFECTIONS: 500mg 3-4
times daily. Children: 30mg/kg/day in 3-4 equally divided doses. IN severe
infections upto 60mg/kg/day may be given. Renal impairment: In patients
with severe renal damage lincomycin doses should be reduced to 25-30 %.
Effective against most Gram-postive organisms. Bacteriostatic or bactericidal depending on sensitivity and concentration. Susceptible organisms unclude Staph. aureus, Staph. albus, beta-haemolytic Streptococcus, Strep. viridans, Diplococcus pneumoniae, Clostridium tetani, C.perfringens, Corynebacterium diphtheriae and C. acnes. Not active against Strep. faecalis, Neisseriae, Haemophilus or other Gram-negative pahtogens
Taking Chloramphenicol, Antidiarrhoeals may decrease the effects of lincomycin.Erythromycin along with lincomycin has synergistic effect .Antidiarrhoeals such as loperamide when taken with Lincomycin may worsen diarrhoea which is an adverse effect of lincomycin.
Adverse
Effect & Precautions
Pain
at injection site, stomatitis, glossitis, angioneurotic oedema,
serum sickness, vertigo, tinnitus, aplastic anaemia. Hypotension
and cardiac arrest after rapid i.v. use. Anorexia, Metallic taste,
oesophagitis, abdominal pain.
Severe Renal and Hepatic disease may increase
serum levels of this drug, increasing the chance of adverse
effects. Patients with Gastric and intestinal disease like colitis
( including colitis caused by antibiotics, or enteritis) may have
an increased risk of adverse effects.
Pregnancy: Safety not established.
Breast Feeding: Best ot discontinue breast feeding.
Man: Use with caution.
