Pharmacokinetics

 


Mefenamic acid is absorbed
from the gastrointestinal tract. Peak plasma concentrations occur about
2 to 4 hours after ingestion. The plasma elimination half-life is
reported to be 2 to 4 hours. Mefenamic acid is extensively bound to
plasma proteins. Small amounts have been detected in breast milk. Over
50% of a dose may be recovered in the urine, as unchanged drug or
conjugates of mefenamic acid and its metabolites.

Indication
& Dosage








Oral

PAINFUL MUSCULOSKELETAL CONDITIONS INCLUDING LOW BACK PAIN, RHEUMATOID
AND OSTEOARTHRITIS, ANKYLOSING SPONDYLITIS, CHRONIC JUVENILE ARTHRITIS,
TRAUMA, DENTAL CONDITIONS:
Acute pain: 500mg followed by 250mg 4 or 6
hours taken with food. Other conditions: 500mg t.i.d. Chorinic juvenile
arthritis: 25mg/kg in divided doses. Antipyretic in children: 4mg/kg.

Action

 



Inhibits the enzyme cyclo-oxygenase and reduces the formation of
porstaglandins. Leukotrienes may also be reduced. Mefenamic acid also
acts as an antagonist at prostagland receptor sites. Analgesic action,
independent of anti-inflammatory activity has been demonstrated for
mefenamic acid.

Interactions

Adverse
Effect & Precaution




As of NSAIDs. Dowsiness,
diarrhoea, rashes (withdraw treatmetn), thrombocytopenia, haemolytic
anaemia, aplastic anaemia, Convulsions in overdosage.

 

Bleeding disorders, aspirin or anti-infalmmatory drug induced allergry,
asthma, hpertension, hepatic or renal or cardiac dysfuntion.

Preganancy: Safety not established.

Breast Feeding: Avoid, because of effects on infant’s cardovascular
system

Man: May be used in reduced dose.
