After oral administration it exhibits inconsistent absorption. Hence it
is administered by intravenous route. It is metabolised in liver.

  

Indication
& Dosage


 


IV

ADENOCARCINOMA, OESOPHAGEAL CARCINOMA, MAMMARY, CERVICAL, GASTRIC AND
LUNG, BLADDER CANCERS, SOFT TISSUE SARCOMAS: Usual dose 10mg/m2 given as
a single dose in a running saline infusion ever 4-6 weeks or 2mg/m2
daily for 5 days. Repeated when bone marrow recovers.

Action

 


Converted to an active metbolite. Inhibits DNA synthesis as also
that of RNA and protein. Myy be selectively toxic to hypoxic cells
because it may generate superoxide and hydoroxyl radicals. Effects are
most marked in the late G1 and early S phase of cell cycle.

Interactions

 


Vinca alkaloids: Acute shortness of breath and severe bronchospams have
occured following the use of vinca alkaloids in patients who had
previously or concurrently received mitomycin.

 

Adverse
Effect & Precaution




Nausea, vomiting, fever, alopecia, pulmonary fibrosis, bone marrow
depression, renal toxicity, sterility, stomatitis, amenorrhoea.



Precaution: Children, GI disorders, CNS disturbance, hepatic or renal
impairment, bone marrow depression. Monitor hepatic, renal and
haematological function.

Pregnancy: Contraindicated.

Breast Feeding: Use with caution.

Man: Use with caution.
