It is absorbed mainly from the stomach and upper part of the small intestine, 60% undergoes hepatic metabolism, it has a half life of 2-8 hrs.
Indication & Dosage
Oral
MILD TO MODERATE PAIN ACUTE AND CHORNIC INFLAMMATORY CONDITIONS SUCH AS
(RA), OSTEOPARTHRITIS (OA), SPONDYLOSIS, ANTIPYRETIC: 300-900mg every 4-6
hours upto max of 4g/day.
JUVENILE RHEUMATOID ARTHRITIS: The only condition for which aspirin cn be
used in children, 80-100mg/kg/day in divided doses.
Action
Aspirin is an analgesic, anti-inflammatory and antipyretic. This action
depends on the inhibition of the enzyme cyclo-oxygenase in the pathway of
prostaglandin synthesis. Reduction of PG reduces inflammation, pain and
fever. Fever is also reduced by an effect on the hypothalamus. Aspirin
inhibits the formation of thromboxane A2 in the platelets. This inhibits
platelet aggregation and coagulation. This action lasts until the enzyme
cyclo-oxygenase is regenerated in the platelets.
Interaction with the following drugs are common:Acetaminophen, Diclofenac, Diflunisal, Etodolac, Fenoprofen, Floctafenine, Flurbiprofen, oral Ibuprofen .
Buffered aspirin with choline and magnesium salicylates, or magnesium salicylate can reduce the effectiveness of certain drugs, especially antibiotics. Aspirin suppositories may cause irritation of the rectum.
