Cefazolin is administered parentrally .It reaches high concentrations in plasma and bile.Cefazolin has a longer half life( 2 hrs )hence used in surgical prophylaxis.80 % is protein bound. 90 % is excreted in the urine.
IV/Deep IM
RESPIRATORY TRACT INFECTIONS, UTI, SKIN AND SOFT TISSUE INFECTIONS, BONE AND JOINT INFECTIONS: 500mg-1g every 8-12 hours. Children: 25-50mg/kg/day in 2-4 divided doses,
SEVERE INFECTION: ENDOCARDITIS, SEPTICAEMIA: 1.0-1.5g every 6 hour.Children: Upto 100mg/kg/day in divided doses.
PERIPERATIVE PROPHYLAXIS: 1.0g 1.2-1 hour before start of surgery followed by 0.5-1.0g every 6-8 hours for 24 hours.
Followed by 0.5-1.0g every 6-8 hours for 24 hours.
NOTE: Renal impairment requires dose modification according to creatinine clearance.
Action
Bactericidal. Gram-positive including Staph. aureus(penicilllinase-producing
strains also), Group A betahaemolytic streptococci, Strep. pneumoniae,
Gram-negative E.coil, proteus mirabilis Klebsiella, Haemophilus
influenzae, Enterobacter.
Probenecid increases the blood level of Cefazolin. Concurrent use of diuretic with Cefazolin may increase the nephrotoxicity.
Adverse
Effect & Precaution
Nausea, epigastric distress,
diarrhoea, vomiting, skin rash, urticaria, genital pruritus,
pseudomembranous colitis and genital moniliasis.
Precautions
are to be taken in Renal disease .
Pregnancy: Use with caution.
Breast Feeding: Use with caution.
Man: May be used.
