Pharmacokinetics

Ceftizoxime is
given parenterally. It is not metabolised in the body and is excreted
through the kidneys. Plasma half life is 1.5-2 hours.

Indication & Dosage

 


SEVERE INFECTIONS: SPETICAEMIA, INTRAABDOMINAL INFECTIONS, THOSE IN
IMMUNOCOMPROMISED PATIENSTS, MENINGTITIS: 2-3g every 8 or 12 hrs. In
life threatening infections 3-4g IV 8th hourly or 2g IV every 4 hours.

INFECTIONS OF RESPIRATORY TRACT, SKIN AND SOFT TISSUE AND BONE AND JOINT
INFECTIONS OBSTETRICAL AND GYNAECOLOGICAL INFECTION;1-2g every 8 or 12
hrs. 

UTI; 0.5 to 1g hourly.

GONORRHOEA; 1g IM single dose.

NOTE: Children; 3 months to 12 yeares; 30-60mg/kg/day in 3 divided
doses. In severe life threatening infections upto 200-300mg/kg/day have
been given. Daily max not more than 6g. Children under 3 months; current
data inadequate. Elderly; reduced doses if renal impairment is present.

Action


 


Active against most Enterobacteriaceae, haemophilus influenzae,
Neisseria gonorrhoeae, Providencia, Vibrio and some strains of
Pseudomonas, Streptococci, Staphylococci, Listeria, monocytogenes,
C.diphtheriae and many anaerobes.

Interactions



Probenicid reduces the renal clearance of Ceftizoxime. Chlorampenicol
reduces the drug’s activity.

Adverse
Effect & Precautions

 


Skin rash, leucopenia, thrombocytopenia, candidiasis, rise in liver
enzymes and blood urea. Rarely nausea, vomiting and diarrhoea. 

 

Precautions are to be taken in renal failure, hypersensitivity to
penicillins, and pseudomembranous colitis.

Pregnancy: Use with caution.

Breast Feeding: Use with caution.

Man: Use with caution.
