Oral absorption is not predictable and bioavailablity is low. Better
effects are produced with IM or IV administration. It is highly bound to
plasma as well as to tissue proteins. Brain concentration is greater
than plasma concentration. It is metabolised in the liver.Plasma half
life is around 30 hours. Acute effects of a single dose usually last for
6-8 hours. It has a cumulative effect on chronic administration, so it
is possible to give the maintanance dose once a day.

Indication & Dosage


 

Oral
SCHIZOPHRENIA, SCHIZO-AFFECTIVE DISORDERS, PARANOID STATES, PANIC-ANIXITY,
MANIA AND HYPOMANIA, AGGRESSIVE BEHAVIOUR: 25-1000mg daily rarely higher
doses upto maximum 2.0g/day. Children: Use with care. 50-200mg/day
depending on weight
IM
FOR ACUTE SYMPTOMS: Adults: 25-50mg 3 or 4 times daily. Elderly: Adult
doses with caution.

A neuroleptic. Acts by blocking the postsynaptic dopamine D2 receptor i
the mesolimbic dopaminergic system.

Interactions
  
Chorpromazine has additive depressant effect with sedatives, hypnotics,
antihistamines, opiates and with alcohol. It potentiates the anti-cholinergic
effects of antiparkinson agents and tricyclic-antidepressants, which may
lead to a anti-cholinergic crisis. It has additive orthostatic
hypotensive effect in combination with MAO inhibitors.

Adverse
Effect & Precaution

Cholestatic jaundice,
drowsiness, lethargy, dry mouth, weight gain, hyptension (on parenteral
admintration) respiratory depression leucopenia, tardive dsyskiness
akathisia, acute dystonia, skin rarely lens opacities, Interfer-Cardiac
arrhythmia, impaired temperature regulation.

 

Precautions are to be taken in CVS disease, pregnancy, lactation and in
elderly patients.
Pregnancy: Use with caution.
Breast Feeding: Use with caution.
Man: May be given in reduced dose.
