completely absorbed and is concentrated in lepromatous skin, liver,
kidney and muscle.

Indication &
Dosage

 


For treatment leprosy. Dermatitis
herpetiformis

Adults : Leprosy 50 to 100mg/day depending upon body wt.

Dermatitis herpetiformis : start with 50mg daily and increase obtained.
Dose reduced to a minimum maintenance level as soon as possible.

Children : 1-2mg/kg/day, start with minimum dose and increase weekly so
that at the end of 7th week patients is receiving max. dose.

Interaction

 

Serum concentrations of
dapsone are increased, with a consequent increased risk of adverse
effects, during concurrent administration of probenecid, probably as a
result of reduced renal excretion of dapsone. Increased dapsone
concentrations have also been reported in patients receiving
trimethoprim, and patients receiving this combination may be at
increased risk of dapsone toxicity. Rifampicin reduces serum
concentrations of dapsone to a level that may compromise efficacy in
infections other than leprosy. Rifampicin concentrations are generally
unaffected. Dapsone may antagonise the anti-inflammatory properties of
clofazimine.

Precaution




Haemolysis is most common and is dose related. Patients with
G-6PD deficiency are more susceptible. Nausea, vomiting, anorexia,
headache, methaemoglobinaemia, drug fever, insomania and paresthesia.
Rarely hepatitis and agranulocytosism lepra reaction.


 


S
pecial Precaution : Initially low dose
is given and then gradually the dose is increased.

Pregnancy : Use with caution.

Brest Feeding : Discontinue nursing or the drug

Men : May be given in reduced dose.
