Pharmacokinetics

Top

    

Nalidixic Acid when given orally is readily absorbed
from the GIT, 80% is eliminated through the kidneys. Both active &
inactive forms of Nalidixic acid is found in the urine.Concentration in
urine is 20 to 50 times that in plasma.

Indication and Dosage

Oral:


BACTERIAL DIARRHOEA, UTI: Adults: 1g q.i.d.for 1-2 weeks. For
long termtherapy, daily dose should not exceed 2g. Children 3 months- 12
years: 60 mg / kg daily in 4 divided doses.

Action

   

Bactericidal action by inhibition of bacterial DNA
gyrase. Acts against Gram-negative bacteria including Proteus spp, E.coli,
Klebsiella, Aerobacter and Shigella spp.

Interactions

Interaction occurs with Anticoagulants :
Patients taking Nalidixic acid with anticoagulants may have an
increased chance of haemorrhage.Nitrofurantoin should not be used
concurrently.

Precautions

Adverse effects : the most important toxicity
is neurological toxicity such as headache, drowsiness, vertigo,
visual disturbances and occasionally seizures.Prolonged use has
produced parkinsonism like symptoms, leukopenia and biliary
stasis. When used in individuals with G6PD deficiency, they may
develop hemolysis.
