Nimesulide is not a classical NSAID, it has weak action against prostaglandin synthesis [cox-2 inhibitor] but has a potent Anti-Inflammatory action.It behaves as a competetive inhibitor of histamine release in-vivo and in-vitro, hence posseses antihistaminic and antiallergic properties.
Oral
PAINFUL INFAMMATORY
CONdDITIONS ASSOCIATED WITH THE MUSCULOSKELETAL SYSTEM, DYMENORRHOES,
THROMBOPHLEBITS, POSTOP AND DENTAL PAIN, ENT INFLAMMATIONS:
Adults usually 100mg b.i.d Children: 5mg/kg/day in 2 or 3 diveded doses.
Action is somewhat different from that of classic NSAIDs. It is a relatively weak inhibitor of prostaglandin synthesis but has potent antiinflammatory activity. It acts as a inhibitor of histamine release, and reduces in vitro, superoxide anion formation by activated neutrophils. May also inhibit release of tumour necrosis factor alpha and thus reduce the formation of cytokines.
When Nimesulide is taken along with Cyclosporine, Digitalis glycosides, Lithium, Methotrexate, Phenytoin there will be elevated serum levels of these drugs and an increased chance of adverse effects may occur.
Those with Alcohol abuse, Colitis, Crohn’s disease, diverticulitis, gastric ulcer, Diabetes mellitus, Hemorrhoids, Hepatitis, renal disease, rectal irritation or bleeding, Systemic lupus erythematosus and Tobacco abuse may have increased chance of adverse effects.
